Effect of Electroacupuncture Combined with Bone Marrow Mesenchymal Stem Cells on Neurotrophic Factors of Rat Models with Diabetic Peripheral Neuropathy

Abstract
Objective:To investigate the effect of electroacupuncture( EA) combined with bone marrow mesen-chymal stem cells ( BMSC) transplanted on the expression of nerve growth factor ( NGF) and neurotrophins-3 (NT-3) in the sciatic nerve of rats with diabetic peripheral neuropathy(DPN).Methods:The rat models with diabetic peripheral neuropathy were established by streptozotocin peritoneal injection method.Healthy SD rats were randomly divided into a model group,an EA group,a BMSC group and an EA+BMSC group.EA treat-ment was applied to“Ganyu”,“Piyu” ,“Shenyu” and“Zusanli” in EA group;BMSC suspension was infused into the caudal vein in BMSC group;EA+BMSC group was combined EA with BMSC.14d after modeling, the conduction velocity of sciatic nerve was measured,and the mRNA expressions of NGF and NT-3 in the sciatic nerve were detected by RT-PCR.Results:Compared with the model group, the conduction velocity of sciatic nerve was significantly increased in EA group, BMSC group and EA+BMSC group (P<0.01),and the mR-NA expressions of NGF, NT-3 in sciatic nerve were significantly increased (P<0.01).Compared with EA group and BMSC group, the conduction velocity was significantly increased in EA+BMSC group, and the ex-pressions of NGF and NT-3 were increased (P<0.05).Conclusion:The combined treatments promote con- <br> duction velocity of sciatic nerve more significantly than either of the single treatment in DPN rats , which may be related to the combined treatments which have synergistic effect on increasing the expressions of NGF and NT-3 .关键词
