Abstract
Objective:To investigate the effect of moxibustion on endogenous neural stem cell differentiation in rats after cerebral ischemia reperfusion,to clarify the mechanism of action of moxibustion in the treatment of cer-ebral ischemic stroke, and to provide theoretical basis for moxibustion therapy in the treatment of ischemic stroke.Methods:72 Wistar rats were prepared by modified thread embolism of middle cerebral artery focal cere-bral ischemia reperfusion model ( MCAO model) ,and they were randomly divided into a sham operation group, a model group and a moxibustion group.Every group was subdivided into 3d, 7d, 14d, 21d 4 time points and at each time point there were 6 rats.The intraperitoneal injection of Brdu was used to label proliferating cells. The ratsˊ“Baihui, Taiyang( double)” points were selected to conduct the experimental study.TTC staining and HE staining were used to understand brain ischemia and pathological brain tissue perfusion changes.Apply im-munofluorescence technique, BrdU/GFAP, BrdU/NSE double staining.Microscopic image analysis system was used to analyze the positive results.Results:In moxibustion group at each time point neural function defect score was compared with that of the model group and the score was decreased with a significant difference ( P <0.05 ) .At the same time point neural function defect score in the moxibustion group was compared with that in the model group,and there was a significant difference ( P<0.05);moxibustion could promote the increase of BrdU/GFAP, BrdU/NSE positive cells after cerebral ischemia injury, reaching the peak after 3 days.There was a significant difference compared with the model group and sham operation group (P<0.05).Conclusion:Moxibustion can significantly promote the recovery of neural function in rats with cerebral ischemia after reperfu-sion;moxibustion can promote the expression of brain tissue of cerebral ischemia and reperfusion BrdU/GFAP positive cells, BrdU/NSE positive cells, showing that moxibustion can promote neural stem cells into neurons and glial cell differentiation.关键词
