靶向干扰CDK2AP1对乳腺癌MCF-7细胞体外迁移和侵袭的影响
CDK2AP1 knockdown inhibits migration and invasion of human breast cancer MCF-7 cell line in vitro
摘要
Abstract
Objective To investigate the role of cyclin- dependent kinase 2- associated protein 1(CDK2AP1) in migration and invasion of human breast cancer cells. Methods Recombinant lentivirus pGCLV- CDK2AP1- shRNA and negative control pGCLV- GFP were constructed and transfected to human breast cancer MCF- 7 cells. Migration and invasion ability of MCF- 7 cells were determined with xCELLigence system and Transwel chamber assay, respectively. Results Recombinant lentivirus pGCLV- CDK2AP1- shRNA and negative control pGCLV- GFP were transfected into MCF- 7 cells successful y. After transfection, expression levels of CDK2AP1 mRNA in MCF- 7 cells were reduced significantly(P<0.05). Compared to control MCF- 7/RNAi- NC cells, MCF- 7/RNAi- CDK2AP1 significantly decreased the migration and invasion ability of MCF- 7 cells. Conclusion Si-lence of CDK2AP1 significantly inhibits migration and invasion of human breast cancer MCF- 7 in vitro.关键词
