鸦胆子素D对人胰腺癌细胞株Panc-1体外增殖的抑制作用
Brucein D inhibits proliferation and induces apoptosis of PANC-1 pancreatic adenocarcinoma cell line
摘要
Abstract
Objective To investigate the effect of brucein D (BD) on proliferation and apoptosis of pancreatic adeno-carcinoma cel s. Methods Human pancreatic cancer PANC- 1 cel s were treated with 0, 5, 10, 20, 30 and 60μmol/L BD for 24, 48 and 72h. The cel proliferation was detected by cel counting kit- 8 (CCK- 8) assay, cel apoptosis was detected using Annexin V- FITC/PI. The protein expressions of Bax and Bcl- 2 were detected using Western blot. Results BD significantly inhibited pro-liferation of PANC- 1 cel s, up- regulated Bax protein expression and down- regulated Bcl- 2 expression in a dose- and time- de-pendent manner(P<0.05). Conclusion BD can significantly inhibit the proliferation and induce apoptosis of PANC- 1 cel s in vit-ro, which is associated with up- regulation the expression of Bax and down- regulation the expression of Bcl- 2.关键词
