HPLC-UV法测定大鼠血清中柴胡皂苷 C浓度
HPLC-UV for the Determination of Plasma Concentration of Saikosaponin C in Rats'Serum

Abstract
Objective:To establish an HPLC method for determining the plasma concentration of Saikosaponin C in rats'serum and to study the pharmacokinetics of Saikosaponin C in rats.Methods:The protein of plasma samples was precipitated with methanol and the plasma concentration was determined by using HPLC.The HPLC procedure was performed on the chromatographic column of Waters C18 (4.6mm ×250 mm,5μm) , and the mobile phase was acetonitrile-0 .1%phosphoric acid solution .The flow velocity was 1 .0 ml/min and the detection wavelength was 210nm.The plasma concentration of Saikosaponin C in rats was determined after sin-gle ip administration of 528 mg/kg,and the data were processed with the pharmacokinetic software DAS 2.0. Results:Calibration curves were linear over 58.4~7300mg/L(r=0.9994) and the LLOD(S/N=3) of Sai-kosaponin C was 14.6mg/L and the average recovery was higher than 85%.RSD of intra-day and RSD of inter-day were all lower than 8%.After single ip administration of 528 mg/kg, the main pharmacokinetic parameters of t1/2z,AUC0-t,MRT0-t were (1.25 ±0.84)h,(943.79 ±39.16)h/(mg・ L),(0.91 ±0.05)h. Conclusion:The method is easy and accurate, which can be used to analyze quantitatively the plasma concen-tration of Saikosaponin C and it is applicable to study the pharmacokinetics of Saikosaponin C in rats.关键词
