Abstract

To investigate the isolating rate of enterohemorrhagic Escherichia coli (EHEC)from cows and their phylogenetic relationships.EHEC were isolated and identified by polymerase chain reaction (PCR) and CT-SMAC separation.PCR was used to detect genes of stx1,stx2,eae,ehxA,saa,wzxO157.All i-solates were detected for the cytotoxicity in Vero cells.The isolation rate of EHEC was 7.1% (40/565). And those 40 EHEC belonged to 17 O serotypes,of which the O157 (4/40),O26 (4/40),O91 (7/40), O100 (4/40),O97 (6/40),O92 (2/40)were common serotypes.The isolation rates of stx1,stx2, ehxA,and saa were 82.5% (33/40),75% (30/40),90% (36/40),(27.5%11/40),respectively,which was higher than the other genes,e.g.eae (5%,2/40),wzxO157 (10%,4/40).All stains which carry stx1/stx2 showed cytotoxic activity in Vero cells.A total of 7 STs were identified in 40 isolates through MLST analysis,and ST297 was the common STs,was 57.5% (23/40).Phylogenetic analysis revealed a closer genetic relationship between our isolates and the EHEC strains which were epidemic in the world.It could be concluded that there exists molecular diversity among 40 EHEC,which showed genetic relation-ship with the popular EHEC worldwide.Except for EHEC O157,non-O157 EHEC strains could also threaten public health of humans,which should be monitored and detected frequently.
