摘要

Abstract

To explore the genetic variation characteristics of porcine epidemic diarrhea virus (PEDV)in re-cent years in China,a continuous RT-PCR was used to amplify the whole spike gene of 4 PEDV field strains.After sequencing and analyzing by some molecular software,there were two insertion sites at ami-no acid sites of 59-62(GENQ)and 140(N)and one deletion site at site of 163-164(NI)in the spike gene of fields stains with 4 161 bp(4 158)length.The phylogenetic tree showed that new field strains are disposed of a different group of G2 with other epidemic strains in recent years,while G1 group consists mostly of vaccines of CV777,DR13 and KPEDV-9 which share an identity of 92.4%-93.3% with field strains of spike gene at amino acid level.Linear B-cell epitope analysis showed that the threshold values at sites of 52-62,360-365 and 752-781 of field stains were high than that of vaccine strains,which indicated an intense relationship with pathogenicity,receptor-binding ability and infectivity.
