Abstract
Aim: To explore the effects of ginsenoside Rd( GSRd) on osteogenic differentiation of high glucose-in-duced human periodontal ligament stem cells(PDLSCs). Methods: PDLSCs were pretreated with 30 mmol/ L D-glucose and 5 mmol/ L D-glucose, then the osteogenic differentiation indexes of PDLSCs such as the ALP activity, mRNA expres-sions of ALP, OCN, and protein expression of OPN were detected. To evaluate the effects of GSRd on oxidative stress of high glucose-induced PDLSCs, the protein and mRNA expressions of SOD and CAT were detected. Results: High glucose treating decreased the ALP activity of PDLSCs(P < 0. 05), but 10 and 40 μmol/ L GSRd could improve the ALP activity of PDLSCs significantly(P < 0. 05). Compared with the control group, GSRd upregulated the mRNA expressions of ALP, OCN, and protein expression of OPN(F = 102. 893, 56. 061, 60. 296, P < 0. 05). GSRd also upregulated the mRNA and protein expressions of CAT and SOD(F = 48. 393, 67. 134, 29. 837, 7. 539,P < 0. 05). Conclusion: GSRd could improve the osteogenic differentiation of PDLSCs and reduce the oxidative stress of PDLSCs with high glucose treatment, and then improve the osteogenic differentiation of PDLSCs.关键词
