Abstract
Objective To investigate the effects of uric acid(UA)on the proliferation of cultured human um‐bilical vein endothelial cells(HUVECs)in vitro and its oxidative stress mechanism .Methods The effects of UA(0 ,4 , 8 ,12 ,16 mg/dL)on the proliferation of HUVECs were observed after 24 hours by MTT assay .The levels of NO , SOD ,MDA ,and ET‐1 in the supernatants were detected by nitrate reductase test ,xanthine oxidase test ,thiobarbituric acid test(TBA) ,and ELISA ,respectively .The effects of uric acid on the intracellular reactive oxygen species(ROS) production in HUVECs were measured by flow cytometric analysis using an 2′,7′‐Dichlorofluorescin diacetate(DCF‐DA)fluorescence probe .The effects of N‐acetyl cysteine(NAC)on UA‐inhibited the proliferation of HUVECs ;the levels of ROS and the levels of NO ,SOD ,MDA ,and ET‐1 in the supernatants of HUVECs were observed .Results The results showed that 8-16 mg/dL UA significantly inhibited the proliferation of HUVECs in time and concentra‐tion‐dependent ways(P<0 .05) .12mg/dL and 16mg/dL UA increased the levels of ET‐1 and MDA significantly(P<0 .05) ,the activities of SOD and the level of NO were significantly decreased(P<0 .01) .N‐acetyl cysteine(NAC) reduced UA‐induced the levels of ROS ;Compared with control group ,the proliferation of HUVECs and the activities of SOD and the level of MDA ,ET‐1 ,and NO were no significant differences .(P>0 .05) .Conclusion UA could obvi‐ously inhibit the proliferation of HUVECs in time and concentration‐dependent ways .Its mechanism may be associat‐ed with uric acid increase the levels of ROS in endothelial cells ,suggesting that uric acid mediated oxidative stress maybe involved in the injury of endothelial cells .关键词
