藏雪莲水提取物对中波红斑效应紫外线辐射后人角质形成细胞中蛋白表达的影响研究
Influence of Saussurea Tridactyla Sch -Bip Water Extracts on the Protein Expression of HaCaT Cells After UVB Radiation

Abstract
Objective To investigate the influence of Saussurea tridactyla Sch -Bip ( SSB ) water extract on the expressions of interleukin 6 ( IL-6 ) , tumor necrosis factor alpha ( TNF -α) , p38 mitogen activated protein kinase (p38MAPK), p53, B cell lymphoma factor 2 ( Bcl-2 ), Bcl-2 associated X protein ( Bax ), cysteine proteinase 3 ( Caspase-3) in HaCaT cell after medium wave ultraviolet erythema effect of ultraviolet rays ( UVB) radiation.Methods HaCaT cells were cultured in vitro and randomly divided into 3 groups: control group, UVB radiation group (30 mJ/cm2, radiation 1 h, UVB group), UVB+SSB group (UVB 30 mJ/cm2, radiation 1 h +SSB 5 g/L) .One hour before UVB irradiation, SSB water extracts were added into the culture medium .18 hours after UVB radiation , cells were collected.The morphology of cells were observed by trypan blue staining .The expressions level of IL-6, TNF-α, p38MAPK, p53, Bcl-2, Bax and Caspase-3 were detected.Results Compared with control group , in UVB radiation group , the number of HaCaT cells stained increased , HaCaT cells appeared swelled and floated , but the cell morphology could still be seen; compared with UVB radiation group , in UVB+SSB group, the number of HaCaT cells stained decreased , HaCaT cells appeared less swelled , and the morphology of cells were largely restored.UVB radiation group was higher ( P<0.05) in the expressions level of IL-6, TNF-α, p38MAPK, p53, Bax, Caspase-3 and lower ( P<0.05 ) in Bcl-2 expression than control group; UVB +SSB group was lower ( P<0.05) in the expressions level of IL-6, TNF -α, p38MAPK, p53, Bax and Caspase-3 and higher ( P <0.05) in the expression level of Bcl-2 than UVB radiation group.Conclusion SSB water extracts can reduce the expressions level of IL-6, TNF-α, p38MAPK, Bax, Caspase-3 and increase the expression level of Bcl-2 in HaCaT cells after UVB radiation.关键词
