摘要

Abstract

Objective To investigate the mRNA expression of TGF-β and FOXP3 during oral carcinogenesis. Meth-ods The tissues were obtained from 4-nitroquinoline-1-oxide-induced animal models with tongue carcinogenesis estab-lished in early study. Specimens were divided into three groups: normal ( n=8) , dysplasia ( n=8) , and squamous cell carcinoma ( SCC, n=8) . The transcriptional levels of TGF-βand FOXP3 were examined by real time quantitative PCR. Results The mRNA level of TGF-βwas gradually increased from normal mucosa to dysplasia and SCC ( 3. 0 ± 1. 6, 5. 2 ± 1. 6, and 13. 6 ± 3. 0, respectively) . The transcriptional level of TGF-βin dysplasia group and SCC group was sig-nificantly higher than that in the normal group ( P=0. 043, P<0. 001, respectively) . The mRNA expression of FOXP3 in normal group was1. 8 ± 0. 8, while 3. 6 ± 1. 0 in dysplasia group, and 6. 1 ± 2. 5 in SCC group. The expression of FOXP3 in the dysplasia and SCC group was also significantly higher than that in the normal group ( P =0. 004, P =0. 005, respectively) . Furthermore, the expression of TGF-β was positively correlated with the level of FOXP3 in the multistage of tongue carcinogenesis ( r=0. 686, P<0. 001) . Conclusion The upregulation of TGF-βand FOXP3 might be associated with disease progression during 4NQO-induced rat tongue carcinogenesis.
