Rapid automated synthesis of 3'-deoxy-3'-[18F] fluorothymidine based on a domestic 18F multi-function module
摘要
Abstract
Objective To providea simple rapid automated method of synthesis of 18F-FLT by using domestic 18F multi-function module for clinical need. Methods: First, Sep-Pak QMA cartridge was rinsed with anhydrous methanol, then the trapped 18F- was eluted into the reaction vessel with tetraethylammonium bicarbonate as phase transfer catalyst. With 3-N-t-butoxycarbonyl-1-[5'-O-(4,4'-dimethoxytriphenylmeth-yl) -2'-deoxy-3'-O-(4-nitrobenzenesulfonyl)-β-D-threopentofuranosy] thymine as the precursor molecule, the 18F-FLT injection was eventually prepared by nucleophilic reaction, hydrolysis, neutralization, and purification withSep-Pak cartridge. Results The automated synthesis of 18F-FLT was successfully completed with the total synthesis time of about 25~30 min. the uncorrected yield was between 10% and 15%, and both the radiochemical purity and chemical purity were greater than 95%. The retention time of 18F-FLT and 19F-FLT on the analytical HPLC was 14.3 min and the Rf value was about 0.7 according to TLC analysis. Conclusion A reliable, simple and efficient method of preparation of 18F-FLT has been developed, which provides an efficient and rapid drying method for 18F-.and through Sep-Pak cartridge purification to obtain 18F-FKT injection of high radioch evical purity and chemical purity.关键词
