谷子天冬氨酸转氨酶基因部分片段的克隆
Cloning of Partial Fragment of Aspartate Aminotransferase Gene in Foxtail Millet
摘要
Abstract
The genomic DNA extracted from young,no-lesion leaves of foxtail millet cultivar Yugu 1 was as template,and a pair of specific primers was designed according to the reported corn aspartate aminotransferase gene sequence to clone aspartate transaminase partial gene sequence of foxtail millet,so as to provide foundation for alte-ring composition and content of amino acids by genetic engineering,further altering nutritional quality of foxtail mil-let.The results showed that:PCR amplification obtained a 750 bp target fragment.After cloning and sequencing,a 742 bp fragment was obtained.After Blast search through public database,a specific conserved domain belonging to aspartate aminotransferase supergene family was found in cloned sequence,which proved that the cloned fragment was aspartate aminotransferase gene.Then a phylogenetic tree based on the gene sequences was constructed,which indicated that part of monocots and dicots could cluster respectively,but foxtail millet,maize and sugar cane,three C4 plants,could not cluster together.This study provided foundation for further revealing the function of aspartate aminotransferase gene of foxtail millet.关键词
