基因工程菌 K88-K99融合蛋白高密度发酵工艺的初步研究

摘要
Abstract
The five kinds of media,LB,improved LB,improved LB+ trace salt solution,improved Minca and 2×YT+10 g/L glucose were screened,and the initial pH of medium were set as 6.0,6.5,7.0 and 7.5 for protein expression.With automatic microorganism growth instrument,OD values of bacterial liquid were measured every half an hour.Finally 2 × YT + 5 g/L glucose and pH7.0 were confirmed for the growth of bacterial protein expression.The optimal induction time was in 3.0 h-3.5 h after culture by bac-terial growth curve,induction time 4.0 h,oxygen control at 30%,for ETEC E.coli to provide basic data for the research of high density fermentation.关键词
