T-RFLP analysis of soil microbial diversity in Humble Administrator's Garden

Abstract
Terminal restriction fragment length polymorphism (T-RFLP) technique was used to analyze microbial diversity in 3 points of sediment and 3 points of soil in Humble Administrator's Garden. The results show that a-mong the sediment bacteria ,T-RFs fragments with the sizes of 181 bp ,211 bp were the dominant bacteria. A-mong soil bacteria ,T-RFs fragments with the sizes of 91 bp ,205 bp were the dominant bacteria. Among sediment fungi, species of each site were not identical. Among soil fungi ,T-RFs fragment with the size of 355 bp was the dominant fungus. Bacterial and fungal diversity of sediment and soil in Humble Administrator's Garden were dif-ferent. Bacterial diversity was significantly higher than that of fungi. This study has provided valuable data for microbial diversity and community structure of Suzhou gardens.关键词
