摘要

Abstract

Objective To investigate the effect of hesperidin on myocardial ischemia /reperfusion injury of myocardial cells induced by autophagy and its possible mechanism .Methods 24 male SD rats were randomly divided into sham operation group, ischemia /reperfusion (I/R) group and hesperidin group with 8 rats in each group.Sham operation group were treated with 0.5% carboxymethylcellulose sodium orally , after 3 days, thoracotomy were performed to expose the left anterior de-scending coronary artery ( LAD) but not ligation , in I/R group and hesperidin group were blocking LAD ’ s flow for 30 min, then with 4 hours of reperfusion to establish the rat myocardial ischemia /reperfusion injury model .After the operation , detec-ted the serum creatine kinase (CK) and lactate dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde ( MDA) and autophagy protein , microtubule associated protein 1 light chain 3 II ( LC3 II) and Beclin1 level in the myocardial tissue.Results Compared to sham operation group , I/R group rats’ serum CK and LDH levels were significantly increased , SOD activity of myocardial tissue were significantly decreased , MDA content significantly increased ( P <0 .05 ) , compared with I/R group, hesperidin group’s serum CK and LDH levels significantly decreased , SOD activity of myocardial tissue was significantly increased, MDA content decreased significantly ( P <0.05).Compared with the sham group, I/R group rats’ myocardial tissue LC3 II and Beclin1 levels increased significantly , compared with I/R group, hesperidin group ’ s index ex-pression levels were significantly lower ( P <0.05).Conclusion Hesperidin can inhibit myocardial ischemia /reperfusion injury induced cardiomyocyte autophagy , and the mechanism is related with hesperidin reduce the oxidative stress .
