Abstract
Objective: To explore the role and mechanism of aspirin in regulating the expression of endometrial receptivity marker gene HOXA10 and embryo implantation.Methods:In this study, Ishikawa cells were treated with different doses of aspirin for indicated time and transfected with siRNAs against HOXA10. hTe expression of HOXA10 mRNA and protein in Ishikawa cells were determined by Real-time PCR and Western blot. Furthermore, the effect of aspirin on embryo implantationin vitro was determined by anin vitro model of BeWo-Ishikawa cell interaction. Results:Compared with the control group treated with DMSO, the expression of HOXA10 mRNA and protein were up-regulated in Ishikawa cells treated with different doses of aspirin. And, the mRNA and protein levels of HOXA10 were highest in Ishikawa cells treated with aspirin at the dose of 10−5 mol/L (P<0.01). Furthermore,aspirin promoted BeWo spheroids attachment to Ishikawa cells, however, the effect of aspirin on BeWo spheroids attachment was inhibited by HOXA10-specific siRNAs in Ishikawa cells.Conclusion: Aspirin may promote embryo implantation through up-regulation of endometrial receptivity marker gene HOXA10.关键词
