Abstract

Objective: To observe the expression of PAR-1and HIF-1α neuronal apoptosis in rats after cerebral hemorrhage. Methods: 90 SD rats were randomly divided into normal control group (n=30), sham operation group (n=30),and ICH group (n=30). Autologous blood was injected into the caudate nucleus of rat to produce cerebral hemorrhage model by using stereotaxie technique. Decapitation was taken at 6hr, 1d, 2d, 3d, 5d and 7d after the model was produced. PAR-1 positive cell, HIF-1α positive cell, and TUNEL positive cell,were detected by immunohisto-chemistry,and VEGF in intracerebral hemorrhage group.Results: There was only a few PAR-1,HIF-1α expression in normal control group,sham operation group and difficultly observated apoptotic cell (TUNEL positive cell). PAR-1 positive cell and TUNEL cell increased in intracerebral hemorrhage group at 6hr, PAR-1 positive cell and TUNEL cell reached a high level at 3d, PAR-1 positive cell descended to normal level at 7d, but TUNEL cell reached a higher level at 7d. HIF-1α was not expressed obviously in intracerebral hemorrhage group at 6h, increased obviously at 1d, reached a high level at 3d, but expressed a higher level at 7d, had statistical significance to normal control group,compare sham operation group (P<0.05). VEGF was expressed obviously in intracerebral hemorrhage group at 6hr, reached a high level at 3d, but a higher level at 7d, statistically different from normal control group,sham operation group (P<0.05).Conclusion:PAR-1, apoptotic cell in the brian hematoma perienchyma markedly increased after ICH;HIF-1αpositive cell,VEGF contents obviously increased.
