Abstract
Objective:To investigate the effect of perifosine on cisplatin-induced apop-tosis and Caspase-3 and PARP expression in human ovarian clear cell carcinoma ES2 cell line. Methods:Human ovarian clear cell carcinoma ES2 cells were treated with different doses of perifosine(0、2、4、6、8 and 10μmol/L) for 48h. CCK-8 assay was used to evaluate the minimal effective perifosine dose. Then the ES2 cells were treated with perifosine at minimal effective dose combined with different doses of cisplatin(0、10、40、70 and 100 μmol/L) . Bürgi formula was used to assess the synergy effect between perifosine and cisplatin. Apoptosis was studied by DAPI staining analysis and flow cytometer. Cellular mitosis was detected by Giemsa staining as-say. The expressions of Caspase-3(cleaved) and PARP(cleaved) were measured by Western blot. Results:Perifosine at 4μmol/L was the minimal effective dose on ES2 cells. Combined treatment of perifosine at 4μmol/L and cisplatin at 40μmol/L showed the best effect in cell growth inhibition ( q=1 . 21 ) . DAPI staining analysis showed that a large number of karyorrhexis and apoptotic bodies were observed in the combination group. The flow cytometer analysis dis-covered that combination of perifosine and cisplatin synergistically enhanced apoptosis of ES2 cells in comparison with perifosine or cisplatin alone (q=1. 32,P<0. 01). Giemsa staining as-say showed that combinated treatment of perifosine and cisplatin significantly inhibited cellular mitosis compared with perifosine or cisplatin alone ( P<0 . 01 ) . Western blot showed that treat-ment of perifosine combined with cisplatin induced increased level of Caspase-3(cleaved) pro-tein and decreased level of PARP( cleaved) protein. Conclusion:Combined treatment of peri-fosine and cisplatin induced apoptosis in ES2 cells through increasing the expression of Caspase-3(cleaved) protein and decreasing the expression of PARP(cleaved) protein.关键词
