Abstract
Objective:To use the Closed-Sheet mehtod to cryopreserve small numbers of human spermatozoa ultra-rapidly,with the aim of finding the best way to cryopreserve. Meth-ods:Normal semen samples from 20 patients were collected at Dalian Municipal women and childrenˊs Medical Center Reproductive Center. Semen samples were prepared by swimming-up for the experiment. In order to find the best volume of droplets for freezing small numbers of hu-man spermatozoa with cryoprotectants,the freezing effect was compared with different volume, respectively 0. 5μl,1. 0μl,3. 5μl. At the same time,we tried to cryopreserve small numbers of human spermatozoa without cryoprotectants,and compared with cryoprotectants group. Results:No significant difference was observed in the sperm recovery rate,motility rate and viability rate among the three groups. No significant difference was observed in the sperm recovery rate,motil-ity rate and viability rate between the spermatozoa cryopreserved without cryoprotectants and with cryoprotectants. Conclusion:The Closed-Sheet method is a highly effective method for cryopreservation of small numbers of human spermatozoa ultra-rapidly,we can obtain a higher recovery rate, motility rate and viability rate after thawing. Cryoprotectants-free has the same cryoprotecting effect as cryoprotectants.关键词
