Abstract

Objective: To investigate the mechanism of amikacin inhibiting platelet agglutination in a case of multi-anticoagulants dependent pseudothrombocytopenia (PTCA). Methods: EDTA-K2 and citrate anticoagulated blood samples were collected from an EDTA dependent PTCA patient. Amikacin was added into blood samples at different time points. Hematology analyzer was used to count platelets, blood smear to observe platelet agglutination, and flow cytometry to de-tect expression of platelet membrane markers, including CD61, CD42b, PAC-1, CD62p. Results: If amikacin was added into the EDTA-K2 anticoagulated blood within one hour after the collection it could inhibit the agglutination of platelet, dissociate the agglutinated platelet without affecting the morphology and distribution of other blood cells, and activation of platelet membrane surface markers CD62p was inhibited ,while the platelet count keep stable for four hours at room tem-perature. With the extension of time, the platelet count decreased obviously in citrated anticoagulated blood. Conclusions:The patient has multi-anticoagulants dependent pseudothrombocytopenia. Addition of amikacin to blood of the patients could normalize platelets count. Amikacin could inhibit platelet agglutination and dissociate agglutination, which may re-late to inhibiting activation of CD62p on platelet membrane by amikacin.
