Inheritance and segregation of microsatellite loci in the hybrid of Oreochromis niloticus♀× Sarotherodon melanotheron♂

Abstract
We measured the inheritance and segregation in the hybrids F1 and F2 of Oreochromis niloti-cus(♀)×Sarotherodon melanotheron(♂) using microsatellite markers. We selected 21 loci with different-sized al-leles in O. niloticus and S.melanotheron from 86 pairs of microsatellites. These loci contained 59 alleles, including 22 specific alleles in O.niloticus and 16 in S.melanotheron. The observed heterozygosity was higher in the F1 (Ho=0.998) than F2 (Ho=0.739) generation. Conversely, the effective number of alleles(Ne=2.48) and polymor-phism information(PIC=0.497) in F2 were similar to those in F1(Ne=2.45, PIC=0.489). Heterozygous alleles were observed at all loci in the F1, whereas we found evidence of allelic separation at F2 loci. Eighteen loci followed strict Mendelian segregation (chi-squared:1>P>0.05), whereas two loci (GM145, UNH003) exhibited segregation distortion (P<0.05) and one locus (GM276) exhibited non-segregation. Among the 18 Mendelian loci, the F2 pos-sessed the similar allelic frequencies for O. niloticus specific alleles (48.3%) and S. melanotheron-specific alleles (47.7%). The genotype frequencies of F1 type, O. niloticus type, and S. melanotheron type observed in the F2 were 58.2%, 19.9%, and 21.9%, respectively. Our results suggest that the F2 grossly inherited the heterozygosity from F1, but that there is also a certain degree of separation.关键词
