Abstract

Objective:To investigate effects of arsenic trioxide (As2O3) on esophageal squamous carcinoma cell line EC-1.Method:The inhibition effect of As2O3(different doses of 0.5,1,2,4,8,16,32,64 μmol/L) on EC-1 cells proliferation at different time points(24,48,72 h) was detected by MTT assay,and the value of IC50 was calculated .Cell cycle change and cell apoptosis were detected by flow cytometric analysis (FCM).Result:As2O3 was able to inhibit the proliferation of esophageal squamous carcinoma cell line EC-1,with the dose of As2O3 increasing and the effect time prolonging,the inhibition of cells proliferation was enhanced.The IC50 in EC-1 cells was 18.74 μmol/L. As2O3 could induced G2/M phase arrest and the reduction of the ratio of cells in G0/G1 and S phases expectively. As2O3 could enhance the radiation-induced apoptosis.Conclusion:The proliefration inhibition action of As2O3 on esophageal cell line Ec-1 depends on the concentration and the contact time. As2O3 hyperthermia can enhance the killing effect of radiation on tumour cells mainly through affecting the EC-l cell cycle, and the induction of apoptosis is the second reason.
