Calycosin-7-O-β- D-Glucoside Transformation into Calycosin by Incubation with the Snailase
摘要
Abstract
Objective To study the best best conditions and methods to conduct the transformation of calycosin-7-O-β- D-gluco-side into calycosin using the enzymatic hydrolysis with snailase. Methods The snailase was dissolved in a suitable water in constant temperature shaking table and then added dimethyl sulfoxide ( DMSO ) which calycosin-7-O-β- D-glucoside was dissolved in, kept the reaction for 30 min, and finally terminated the reaction with acetonitrile. Results When the snailase and calycosin-7-O-β- D-glucoside were in the ratio of 1:2, added with 200 mL water ( keeping in 35-37 ℃) , added calycosin-7-O-β- D-glucoside that was dissolved in 1 mL of DMSO, and the pH was maintained at between 5-8, then quenched with an equal amount of acetonitrile af-ter incubating 0. 5 h. Conclusion This method can easily transform the calycosin-7-O-β- D-glucose into calycosin ( ﹥ 98%) , and the operating process is simple and effective.关键词
