Abstract
Objective To detect the effect of Castration-Resistant Prostate Cancer PC3 cells with trip⁃tolide(TPL) alone or combined with Bortezomib(BZM). Methods MTT assay was performed to evaluate the growth in inhibitory rate of PC3 cells.PC3 cell apoptosis were determined by flow cytometry.RT-PCR was used to analyze the mRNA expressions of vascular endothelial growth factor(VEGF). Results TPL and BZM both inhibited the growth of PC3 cells in a time-and-dose dependent manner,and the single drug of TPL was stron⁃ger than BZM(P<0.05)in the same concentration.The combination group was stronger than the TPL or BZM alone(P<0.05).From flow cytometry resuLts, the combination group was also stronger than the TPL or BZM alone(P<0.05). RT-PCR showed that TPL and BZM both down-regulated the mRNA expression of VEGF (P<0.05), combination was more obvious than the single drug. Conclusion TPL and BZM alone or combi⁃nation inhibit the growth of PC3 cells and induce apoptosis .There is a synergistic effect when combination of TPL with BZM is in use.关键词
