Cloning and Expression Analysis of Interleukin-6 in Grass Carp (Ctenopharyngodon idella)

Abstract
Interleukin 6 (IL-6) is a pleiotropic cytokine that plays major roles in regulating immune responses, acute phase reactions haematopoiesis and inflammation. The IL-6 cDNA sequences were cloned from the head kidney of Ctenopharyngodon idella by RT-PCR and Smart RACE methods. Results showed that the full-length of cDNA of Ctenopharyngodon idella (CiIL-6) was 1 145 bp, containing an open reading frame of 702 bp, encoding 233 amino acids with an estimated molecular weight of 26.74ku and an estimated isoelectric point of 8.27 including a 5'-terminal untranslated region (UTR) of 86 bp, a 3'-terminal untranslated region (UTR) of 357 bp and an open reading frame (ORF). Amino acid homology analysis showed that the CiIL-6 amino acid homology with the zebrafish( Danio rerio) IL-6 up to 76%, which was the closest genetic relationship, but less than 30% homology with other species. The expressions of IL-6 in various tissues of healthy grass carp were analyzed by semi-quantitative reverse transcription PCR technology. The results were demonstrated that in the Thymus, head kidney and spleen had the highest expression of CiIL-6, and in the gills, stomach and heart had the lowest expression.关键词
