Abstract
Objective To study the effects of different concentrations of serum in culture medium on biological characteristics of bone marrow stromal cells( MSCs) in mice.Methods Bone marrow stromal cells of mice were cultured by whole bone marrow culture method.The primary cells were cultured using the mediums containing the serum with the concentrations of 10%( Group A) ,15%( Group B) and 20%( Group C) ,respectively.The morphology and proliferation of the cells were observed.Results MSCs proliferated rapidly in the mediums containing the serum with the concentration of 10%,and the cells with shuttle shape were common.In the mediums containing the serum with the concentration of 20%,the areas of the cells increased and proliferated slowly.After 7 days of culture,the sum of the cells and the proportion of shuttle-shape cells in Group A were (6.0 ±0.2) ×108/L and (86.3 ±3.2)%,respectively,which were more than those in Group B or Group C(P<0.05).The proliferation duration for increasing 80%in Group A was (7.1 ±0.6)days,which was less than that in Group B or Group C(P<0.05).And the duration for cell senescence in Group A was (144.±1 .2)days,which was more than that in Group B or Group C(P<0.05).Conclusoi n For MSCs culture in vitro in mice,the appropriate concentration of serum in medium is10 %.关键词
