摘要

Abstract

Objective Using three method of fluorescence quantitative PCR ,PCR‐LDR and direct sequencing . we compared the genotyping of UGT1A1 * 28 ,UGT1A1 * 6 ,UGT1A1 * 93 and UGT1A1 * 60 four loci of UGT1A1for 45 cases of peripheral blood of tumor patients .Methods The fluorescence quantitative PCR ,PCR‐LDR and direct sequencing were used to detect the polymorphisms of UGT1A1 gene four loci .Results It's consistent for the result of four loci genotyping in 45 patients by the three methods .Conclusion Three detection methods have ad‐vantages and disadvantages respectively .We can choose the appropriate method according to the specific needs of the experiment .Among them ,with the characteristic of high detection sensitivity ,time saving ,simple operation and avoi‐ding the pollution the method of fluorescence quantitative PCR is suitable for clinical detection .
