成人慢性免疫性血小板减少性紫癜患者血小板G蛋白偶联受体的研究
Study on expression of G-protein coupled receptors of platelets in adult patients with chronic immune thrombocytopenic purpura

Abstract
Objective: To investigate the expression of G-protein coupled receptors of platelets in adult patients with chronic immune thrombocytopenic purpura (ITP). Methods: Adenosine diphosphate (ADP) receptor (P2Y1 and P2Y12),α2A-adrenergic receptor (α2A-AR) and thromboxane A2 receptor (TXA2-R) on platelets were detected by flow cytometry, and Gα protein, proteinase activated receptor 1 (PAR-1) and proteinase activated receptor 4 (PAR-4)were analyzed by Western blot. Results: Flow cytometry showed that the mean fluorescence intensities (MFI) of P2Y1, P2Y12, α2A-AR and TXA2-R expressed in platelets of patients with chronic ITP were 31.4±2.2, 29.6±2.1, 25.8±2.9 and 39.8±3.1, respectively, much higher than those from healthy volunteers (P<0.01);MFI of those in healthy volunteers were 7.8±0.8, 7.2±1.3, 9.8± 0.9 and 4.7±0.6, respectively. Western blot analysis showed that the integrated optical density (IOD) of Gαprotein, PAR-1 and PAR-4 expression in platelets of patients with chronic ITP were 1 046.3±159.96, 832.98±98.81 and 1 518.80±272.45, respectively, significantly stronger than those in healthy volunteers (P<0.01); IOD of those in healthy volunteers were 254.49 ±39.51, 203.92 ±27.47 and 431.27 ±41.86, respectively. Conclusions: Expression of G-protein coupled receptors is increased in platelets of patients with chronic ITP, which indicates that platelets of chronic ITP might take part in the complicated biological process by means of the G-protein coupled receptors signal transduction pathway.关键词
