摘要

Abstract

The studyaims to clarify the relationship between mitochondrial pathway and apoptosis of K562 cells induced by FTY720. Human leukemia ( K562 cells) were cultured in vitro and treated with different concentrations of FTY720 for 24 h, and then survival rate of K562 cells was detected by trypan blue staining method; apoptosis and mitochondrial function were tested by flow cytometry and expression of Bcl⁃2 and Bax related to mitochondria was detected by Western blotting. K562 cells induced by FTY720 for 48 h were collected and cell cycle was measured by flow cytometry. Through the study, we found that with the increase of concentration of FTY720, survival rate of K562 cells was decreased, apoptosis was increased, mitochondrial membrane potential was de⁃creased , and expression of apoptosis promoting protein Bax was significantly increased . Cell cycle a⁃nalysis showed that percentage of SubG1 phase cells was increased while S phase and phase G2M cells were decreased. The study showed that mitochondrial pathwaywas were involved in apoptosis of K562 cells induced by FTY720.
