摘要

Abstract

Objective To establish human ovarian cancer skov3 cell line stably expressing miR-183. Methods The gene of Pre-miR-183,has-miR-183 inhibitor were cloned into eukaryotic expression vector, respectively,yielding pEZX-MR03-183 mimics,pEZX-AM03-183 inhibitor.The pEZX-MR03-183 mimics, pEZX-AM03-183 inhibitor were transfected into 293T cells for acking lentivirus,and the lentivirus transfected in human ovarian cancer skov3 cells.Screening with puromycin and hygromycin B,there got the stable transfection skov3 cell line,and through RT-PCR to verify the expression of miR-183 at transcription level. Results The monoclonal skov3 cell line with stable high and low expression of miR-183 were successfully isolated and cultured. Conclusion The study successfully establish the stably transfected skov3 cell lines of miR-183 that could be used to be an effective cell model to research the biological functions of miR-183.
