红参提取物对血管内皮细胞分泌的影响
Mechanism of Red Ginseng extraction on secretion of endothelial cells

Abstract
Objective To investigate the mechanism of Red Ginseng ex-traction ( Hongshen injection , HSI) on nitric oxide ( NO) ,endothelin-1 (ET-1) and 6-keto-prostaglandin (6-K-PGF1α) released by hu-man umbilical vein endothelial cells .Methods Cultured human umbili-cal vein endothelial cells EA .hy926 which have been randomly divided into five groups , blank group , control group , and three doses ( 40 , 20 , 10 mL · L-1 ) test groups .Tested of the content of NO both by using Griess kit.Then the content of ET -1 , endothelial nitric oxide synthase ( eNOS) , cyclooxygenase-1 ( COX-1 ) and 6-K-PGF1αin the cell supernatant were detected by enzyme linked immunosorbent assay ( ELISA).Results Low and middle dose of HSI could increase the con-tent of NO ( P<0.05 ) .Three dose HSI enhance the eNOS expression level ( P<0.05 ) .Middle and high dose HSI reduce the content of ET -1 in cell supernatant which is similar with the effect of Sildenafil .The 20, 40 mL· L-1 HSI can reduce the content of ET -1 in cell superna-tant ( P<0.05 ) , but has no obvious effect on the content of COX -1 and 6-K-PGF1α( P>0.05 ) .Conclusion HSI can relax the blood vessel by promoting the NO synthesis and release , reducing ET-1 , the effect is based on the NO pathway by increasing the expression of eNOS but not on the prostacyclin I 2 ( PGI2 ) pathway.关键词
