Abstract
Objective To analyze the role of endoplasmic reticulum stress PERK-ATF4-CHOP pathway in matrine induced retinoblastoma cells apoptosis. Methods Different concentrations of 0, 1. 0, 2. 0, 4. 0 g/L matrine was used to treat the HOX-Rb44 cells for 24 h,the cell viability was determined by MTT assay, HOX-RB44 apoptosis situation was determined by ow cytometry,western blot analysis was used to dectect the Bax,Bcl-2,GRP78,GRP94,AFT4,peIF2α and CHOP expression. Results As the increase of concentration of ma-trine,HOX-Rb44 apoptosis rate increased significantly ( P < 0. 05 ) , GRP78 and GRP94 increased significantly, and ATF and PERK downstream peIF2α and CHOP were also increased significantly. Conclusion Matrine could promoted HOX-Rb44 cell apoptosis, endo-plasmic reticulum stress, where PERK-ATF4-CHOP pathway may play an important role.关键词
