Effect of tumor suppressor gene PTEN on the occurrence and angiogenesis of endometri-osis

Abstract
Objective:To study the effect of phosphatase gene ( PTEN) protein on the occurrence and angiogenesis of endometriosis ( EMs) in vivo and in vitro experiments. Meth-ods:Lentiviral vector was used to overexpress PTEN and silence PTEN expression in human primary endometrial cells. Three-dimensional culture in Matrigel gel was used to observe vessel growth mimicry in different PTEN expression groups. SCID mice was used to construct human-mouse chimeric endometriosis model. HE and immunohistochemistry technology were used to detect the pathologic change of ectopic endometrial tissues and the expression of VEGF protein in human-mouse chimeric endometriosis mice model in different PTEN expression groups. Re-sults:Compared with high expression group of PTEN,the cells with low expression of PTEN was easier to form vasculogenic mimicry in Matrigel gel, and angiogenesis significantly increased. Meanwhile,endometrial tissue with low expression of PTEN was easier to form ectopic focus in SCID mice,and VEGF expression was relatively high. Conclusions:PTEN has a certain inhibi-ting effect on the occurrence and development of endometriosis by regulating angiogenesis,so we conclude that PTEN gene can be used to treat endometriosis.关键词
