黄芪多糖和三七总皂苷配伍对糖尿病大鼠肾组织Ⅳ型胶原及层黏连蛋白表达的影响
Effect of Astragalus Polysaccharides Combined with Panax Notoginseng Saponins on the Expression of Col-Ⅳ and LN in Renal Tissues of Diabetic Rats

Abstract
Objective To investigate the effect of astragalus polysaccharides (APS) combined with panax notoginseng saponins (PNS) on the expression of Col-IV and LN in renal tissues of diabetic rats. Methods Diabetic rat model was induced by injecting streptozotocin intraperitoneally. The diabetic rats were randomly divided into the model, APS, PNS, APS+ PNS, benazepril groups. The rats were administered medicine intragastrically one time a day for 8 weeks. Urinary protein level in 24h, blood sugar, blood creatinine and urea nitrogen were determined. The protein expression of transforming growth factor beta 1 (TGF-β1), platelet-derived growth factor (PDGF-BB), type IV collagen (Col-IV) and laminin (LN) in the renal tissue were determined by immunohistochemistry. Results Compared with the normal control group, unrinary protein level in 24 h, blood glucose, creatinine and urea nitrogen level in rats in the model group were significantly higher than those in normal control group (P<0.01); The protein expression of TGF-β1, PDGF-BB, Col-IV and LN in the renal tissue were enhanced (P<0.01). Compared with the model group, unrinary protein level in 24 h, blood glucose, creatinine and urea nitrogen level were reduced (P<0.01); The protein expression of TGF-β1, PDGF-BB, Col-IV and LN in the renal tissue in rats were decreased significantly (P<0.01); The effects of APS combined with PNS were better than those of APS and PNS alone groups (P<0.05, P<0.01). Conclusion The combination between APS and PNS had protective effect on kidney of diabetic rats. Its mechanism may be related to inhibiting extracellular matrix synthesis by downregulating the protein expression of TGF-β1, PDGF-BB, Col-IV and LN.关键词
