摘要

Abstract

Laboratory always obtain the content of algae volume by measuring the chlorophyll a fluorescence intensity. According to the settlement of algae cells, fluorescence value are greatly influenced by static duration, when the static duration is 1min, attenuation rate of fluorescence was ΔF/F0=-9�96% and the determination of algae density is not exact. In the article, a method of algae culture preparation was established by adding agar powder(0�08~0�10g/100mL) to the culture. The suspensibility and stability of the culture was enhanced dramatically by this method, after standing 1 hour, attenuation rate of fluorescence was ΔF/F0=-4�82%≤± 5%�The method enhance the accuracy of chlorophyll contents measurement by chlorophyll spectrofluorimetry and reduces the effect of culture sedimentation to the measurement.
