重组人β防御素-3表达条件的优化及其对抑菌活性的影响
Optimization of the Expression of Recombinant Human Beta Defensin-3 in Pichia Pastoris and Its Effect on Antibacterial Activity
摘要
Abstract
To study the effect on expression level and antibacterial activity of recombinant human beta defensin-3 (rhβD-3)by Pichia pastoris ,the requirements for the flask-shaking culture fermentation,such as pH,culture temperature,methanol concentration and so on,were optimized.The results showed that the optimal expression conditions were as follows:the pH value of the culturing BMMY medium was 6.0, medium volume was 50 mL/500 mL,the yeast was cultured under 28 ℃,290 r/min for 48 hours,and the methanol concentration was 0.5% per 24 hours.It was found that the expression level of rhβD-3 could be increased into 340 μg/mL and the antibacterial activity of rhβD-3 could also be increased.关键词
