栀子苷预处理对 H9C2心肌细胞缺氧/复氧损伤的影响及机制研究

Abstract
Objective] To explore the effects and mechanism of geniposide pretreatment on H9C2 cardiomyocytes during hypoxia/reoxygenation injury. [Methods] H9C2 cardiomyocytes were cultivated in vitro and the model of hypoxia/reoxygenation injury was established. The cells were randomly divided into 3 groups: the control group, hypoxia/reoxygenation injury group and the geniposide pretreatment group. The contents of CK, LDH, MDA and the activity of SOD were evaluated by spectrophotometric method. The cell viability was measured with MTT. The apoptosis rate was detected by Annexin V-FITC/PI staining. [Results] Compared with the hypoxia/reoxygenation injury group, the geniposide pretreatment group can reduce the concentration of CK, LDH and MDA, improve the activity of SOD in myocardial tissue, improve the cell viability and reduce the apoptosis rate. [Conclusion] The geniposide has protective effects on hypoxia/reoxygenation injury, which are associated with scavenging free radical, enhancing the capacity of antioxidation and inhibiting cadioocyte apoptosis.关键词
