金匮肾气丸对创伤后应激障碍孕鼠子代行为及血清5-羟色胺水平的影响研究
Effect of Jinkui Shenqi Pills on the Behavior and Serum Serotonin Level of Posttraumatic Stress Disorder Pregnant Rat Offspring

Abstract
Objective To observe the effect of Jinkui Shenqi pills on the behavior and serum serotonin(5-HT)level of offspring from ″ injured fetus″ of pregnant rats with posttraumatic stress disorder(PTSD). Methods SD rat about 90 days with the proportion of female: male 2: 1 were caged to mate randomly from July 2014 to March 2015. By random number table method,36 female rats that had the first successful copulation were divided into three groups including blank group,PTSD group and traditional Chinese medicine(TCM)group with 12 rats in every group. The pregnant metaphase(gestation 7 - 13 d)rats in PTSD group and TCM group were made into PTSD models. The rats in blank group and PTSD group were commonly fed with conventional feeds on normal experiment condition. The rats in TCM group were made into models and given Jinkui Shenqi pills in the meantime until delivery. Body length and tail length were measured within 24 h of rat birth. On the 30 day after birth,rats were given elevated plus - maze test and the percentage of opening arms enter times(OE% )and the percentage of opening arms standing time(OT% )were calculated. Femoral arterial blood of the rats was sampled on the 30th day after birth,and serum 5-HT level was detected by ELISA. Results There wers significant differences in body length,tail length,OE% ,OT% and serum 5-HT level of rats among the three groups(P < 0. 05);the body length,tail length of rats on the day of birth and OE% , OT% and serum 5-HT level of rats on the 30th day of birth in PTSD group were lower than those of rats in blank group and TCM group(P < 0. 05). No significant differences existed in the body length and tail length of rats on the day of birth and OE% , OT% and serum 5-HT level of rats on the 30th day after birth between blank group and TCM group(P > 0. 05). Conclusion Jinkui Shenqi pills can correct growth retardation and behavioristics change of the offspring induced from ″ injured fetus″ of PTSD pregnant rats,and alleviate stress disorder by reducing 5-HT level.关键词
