β-catenin 介导蛋白激酶 D2 对脑胶质瘤细胞增殖的影响
Involvement of β-catenin in effect of protein kinase D2 on glioma cell proliferation

Abstract
Objective To study whether protein kinase ( PKD2 ) affect glioma proliferation by regulating β-catenin.Methods The effect of PKD2 or β-catenin down-regulation on the proliferation of glioma cells was detected by EDU incorporation assay .Western blotting was applied to detect totalβ-catenin expression level and β-catenin level in each subcellular fraction ,such as the membrane, nuclear and cytosolic fractions of the cells after down-regulation of PKD2. Results Compared with the control group , the proliferation of glioma cells decreased in PKD 2 down-regulation group ( P<0 .01 ) .The total β-catenin protein level did not change , but the cytosolic fractions increased , while those in the nuclear fraction decreased after PKD 2 down-regulation ( P<0.01).Conclusion Down-regulation of PKD2 may inhibit glioma cell proliferation by decreasing β-catenin level in the nuclear fraction .关键词
