辛伐他汀对大鼠成骨细胞骨形成蛋白2及其信号传导蛋白Smad1和Smad5表达的影响
Effects of simvastatin on bone morphogenetic protein-2 and the signal transduction protein(Smad1/5) expression in the osteoblast of rats
摘要
Abstract
Objective:To investigate the possible mechanism of simvastatin ,a cardiovascular system a‐gent ,which has been found a promotive effects in bone formation by inducing the differentiation and proliferation of osteoblast ,involving bone morphogenetic protein‐2(BMP‐2) ,and its signal transducer :sekclsky mothers against dpp 1 and 5 (Smad1 ,and Smad5) .Methods :A continuous tissue cultivation method was used to separate and culti‐vate primary osteoblasts from cranium of SD neonate rats .The relative mRNA expression of BMP‐2 ,Smad1 and Smad5 of osteoblasts were determined by reverse transcription PCR(RT‐PCR) after being exposed to simvastatin [0 (S0) ,10‐8 (S1) ,10‐7 (S2) ,5 × 10‐7 (S3) ,10‐6 (S4) mol/L] for 72 hours .Results :The relative expression of BMP‐2 , Smad1 and Smad5 was significantly increased in a dose‐dependent manner after being treated with simvastatin (10‐8‐10‐6 mol/L) .The 10‐6 mol/L simvastatin showed the best promotive effects on the expression of BMP‐2 ,Smad1 and Smad 5 .Conclusion:The increased expression of mRNA of BMP‐2 ,Smad1 and Smad5 may be involved in the possi‐ble mechanism of bone formation induced by simvastatin .关键词
