Abstract
Objective:To investigate the expression of mTOR/ p70S6K signaling pathways in epithelial ovarian cancer and their significance.Method:120 surgical specimens in Tianjin Center Maternity Hospital from January 2011 to June 2011 were selected,they included the normal ovarian group(group Ⅰ),the epithelial ovarian benign ovarian tumor group(group Ⅱ),epithelial ovarian cancer(group Ⅲ),40 casess in each group. The expression of mTOR,p70S6K in epithelial ovarian tissue was detected by immunohistochemical staining.The expression of mTORmRNA,p70S6KmRNA were detected by Reverse transcription(RT)-PCR method.Using Kaplan Meier and log-rank test of single factor analysis,the mTOR,p70S6K protein expression level(negative and positive group) of ovarian cancer patients’ survival rate were compared.Result:Immunohistochemical staining showed that mTOR,p70S6K expressed in ovarian tissue of group Ⅰ was very low,the expression of ovarian tissue in group Ⅱ followed by,and it was highly expressed in ovarian tissue of group Ⅲ.The expression of mTORmRNA and p70S6KmRNA in group Ⅱ,Ⅲ were significantly higher than those in the group Ⅰ,and the expression of mTORmRNA and p70S6KmRNA in group Ⅲ were significantly higher than those in the group Ⅱ.Nonparametric analysis showed that the expression level of mTOR,p70S6K in epithelial ovarian cancer were correlated with the surgical pathologic stage(P=0.039,0.022) and pathologic grade(P=0.047,0.037),had nothing to do with patients’ age,pathological type,peritoneal metastasis.Single factor survival analysis showed that mTOR, p70S6K protein expression of total survival time in negative and positive,the difference was statistically significant (P=0.021).Conclusion:The higher expressions of mTOR/ p70S6K signaling pathways are related to occurrence, development,metastasis and prognosis of epithelial ovarian cancer.关键词
