酶解-连续流动法在烟梗果胶含量测定中的应用

Abstract
In order to provide a reference for determining pectin in tobacco stem exactly,the applica-bility for the determination of stem pectin content by Hydrolysis - continuous flow method was studied, meanwhile the hydrolysis -continuous flow method was developed,and the enzymatic hydrolysis conditions were optimized for 15 mg of pectinase at 55 ℃ for 3 h in 60mL of sodium citrate /citric acid (pH4.0)buffer solution by single factor experiments during the research.Under this condition,the method has good linearity with a correlation coefficient of 0.999 9,while the recovery of pectin in tobacco stem ranged from 97.8% to 100.3%,and the RSD was 1.06% to 2.43% intraday,1.87% during the day.It showed that the method was satisfactory and could be used for the determination of pectin in tobacco stem simultaneously.关键词
