Abstract

Objective:Comparing the expression of placenta proteins in women with preeclampsia and normal pregnancy with labeled relative and absolute quantification ( iTRAQ) technology and liquid chromatography tandem mass spectrometry ( LC-MS/MS ) technology to screen out the potential plasma biomarkers,and to explore the possible mechanism of them in the development of pre-eclampsia. Methods:From Dec. 2014 to Jun. 2015 the women with se-vere pre-eclampsia and normal pregnancy were collected from the clinic among the pregnancy women who diagnosed and regular checked in Affiliated Hospital of Qingdao University for this study,30 cases in each group. Additionally the normal full-term pregnant women ( control group) were collected as control group. Placenta was collected from women who accept the ce-sarean operation, and protein was extracted from placenta. Afterwards, albuminous, degenera-tion,reduction and enzymolysis were performed. iTRAQ was adopted to achieve the expression difference. Results:The difference proteins in placenta which could be effectivity identified by iTRAQ combined with LC-MS/MS technology were 234,of which 24 proteins were identified in PE group more 1. 5 times(increase ratio>1. 50 or lower ratio<0. 67) than the control group of which 14 points up regulation of preeclampsia,and the other ten points down regulated. Conclu-sion:iTRAQ combined LC-MS/MS technology can effectively filter out the difference proteins between preeclampsia and normal pregnancy.
