糖尿病与氧化应激在颈动脉粥样硬化大鼠内皮功能障碍中的作用研究
Impact of Diabetes and Oxidative Stress on Endothelial Dysfunction of Carotid Atherosclerosis Rats

Abstract
Objective To observe the impact of diabetes and oxidative stress on endothelial dysfunction of carotid atherosclerosis ( CAS) rats.Methods From June to October in 2014, 30 healthy male SPF-grade Wistar rats were divided into control group, CAS group and diabetic CAS group with 10 rats in each group by random table method.CAS group and diabetic CAS group were given intraperitoneal injection of a single dose of Vitamin D3 by 600, 000 U per kg body weight with high-fat diet.In the fifth week, diabetic CAS group was given intraperitoneal injection of a dose of Streptozocin by 45 mg per kg body weight.Control group was fed with basic diet.After 15 weeks, the rats were sacrificed, and 3 mm fresh carotid arterial ring was sectioned from each of the separated carotid arteries.The carotid artery ring tension was measured by adding different concentrations of acetylcholine (10-9 , 10 -8 , 10-7 , 10-6 , 10-5 and 10 -4 mol/L) into HV-4 out body tissue organ constant temperature perfusion system.Carotid artery tissue was observed by Hematoxylin and Eosin staining, and the average optical density of CyPA, KLF2 and eNOS was measured by immunohistochemistry SP method.Results After the addition of different concentrations of acetylcholine into HV-4 out body tissue organ constant temperature perfusion system, the carotid arterial rings of all the three groups had relaxant responses.CAS group and diabetic CAS group were lower than control group in carotid arterial ring tension when acetylcholine was given by 10 -9 , 10-8 , 10 -7 , 10 -6 , 10 -5 and 10-4 mol/L ( P <0.05 ) .Diabetic CAS group was lower than CAS group in carotid arterial ring tension when acetylcholine was given by 10-8 , 10-7 , 10-6 , 10 -5 and 10-4 mol/L ( P <0.05 ) .HE staining showed that carotid artery intima of control group was complete and smooth with endothelial cells and smooth muscle cells in alignment, elastic fibers continuous without breakoff, and no hyperplasia.In CAS group and diabetic CAS group, intima breakage, formation of medial calcification plaques and loss of elastic layer continuity were observed.CAS group and diabetic CAS group were higher in the average optical density of CyPA and lower in the average optical density of KLF2 and eNOS than control group (P<0.05) .Diabetic CAS group was higher in the average optical density of CyPA and lower in the average optical density of KLF2 than CAS group ( P <0.05 ) .Conclusion Diabetes and oxidative stress participate in the formation of endothelial dysfunction of CAS rats, and the mechanism may be related with the increase of CyPA expression.关键词
