Abstract

In the present study, the reverse genetics systems of A/duck/Zhejiang/690/2009(H7N3) (ZJ690) and A/duck/Zhejiang/766/2009/(H7N3) (ZJ766) were established. After 8 plasmids were co-transfected into 293T cells, two viruses were rescued and named as R-ZJ766 and R-ZJ690. Sequence analysis confirmed no unexpected mutations in the genome of two rescued viruses. To test the pathogenecity of wild-type and rescued viruses in mouse, each of 10 six-week-old BALB/c mice were inoculated intranasally with 106EID50 of viruses. Four viruses (ZJ690、ZJ766、R-ZJ690、R-ZJ766) did not kill any mice but all viruses caused weight loss. All viruses were recovered only from the nasal turbinates and lungs of the inoculated mice, suggesting the rescued viruses possessed the same biological characteristics as wild-type viruses. The establishment of the reverse genetics system of two H7 viruses would support the further study on the pathogenic and interspecies transmission of H7 Avian influenza virus.
