Role of caspase-3 and DIAPH3 in invasive breast cancer
摘要
Abstract
Objective To study the possible association of the expressions of caspase-3 and diaphanous related formin 3 (DIAPH3) protein with clinicopathological features and prognosis in invasive breast cancer. Methods The expressions of caspase-3 and DIAPH3 protein in invasive breast cancer tissues from 240 cases and para-cancerous tissues from 44 cases were detected by immunohistochemical EliVision method, and then their relationships with the clinicopathological features and prognosis were analyzed. Results The positive-expression rates of caspase-3 and DIAPH3 protein in the invasive breast cancer tissues (36.7% and 40.8%respectively) were significantly lower than those in the para-cancerous tissues (81.8% and 86.7% respectively, P<0.05). The expression of caspase-3 was negatively correlated with the number of lymph node metastasis, tumor size, advanced tumor clinical stage, vascular tumor embolus and positive expression of P53. The expression of DIAPH3 was also negatively correlated with the number of lymph node metastasis, vascular tumor embolus and advanced tumor clinical stage. Kaplan-Meier single factor analysis showed that the 5-year survival rates in the patients with positive expressions of caspase-3 and DIAPH3 were 95.5% and 95.8% respectively, sig-nificantly higher than those in the negative-expression patients (85.5% and 84.5% respectively). The Cox model analysis showed that the negative expression of DIAPH3 might be a risk factor for the prognosis of invasive breast cancer ( OȒ =1.687, 95%CI: 1.032, 2.758). Conclusions The negative expressions of caspase-3 and DIAPH3 protein are important for the development and prognosis of invasive breast cancer, but there is no correlation between them. The low expression of DIAPH3 is probably one of the independent factors of prog-nosis in breast cancer.关键词
