白念珠菌 FLO8基因突变株构建及鉴定
Construction and identification of Candida albicans FLO8 mutations
摘要
Abstract
Objective To construct Candida albicans FLO8 mutations.Methods We inserted Candida albicans FLO8 gene under the control of the ADH1 promoter in pCP20 plasmid,and plasmids containing the point mutations FLO8 R209T , FLO8 A3 1 1 T ,FLO8 6 54Ter ,FLO8 G723R and FLO8 T7 5 1 D were constructed using site-directed mutagenesis from plasmid pCP20.Then we introduced the inserts with FLO8 point mutations into Candida albicans SN1 52 flo8/flo8 at the endogenous ADE2 gene loci by homologous recombination.Results The plasmids with FLO8 point mutations were constructed successfully by DNA sequencing;The inserts with FLO8 point mutations were exactly recombined to the ADE2 gene loci of Candida albicans SN1 52 flo8/flo8 as confirmed by PCR.Conclusions Candida albicans FLO8 mutations can be efficiently constructed with pCP20 plasmid by site-directed mutagenesis,homologous recombination,etc.关键词
