Fgl2基因过表达对自身免疫性心肌炎大鼠心功能的影响
Effects of Fgl2 gene over expression on heart function in autoimmune my-ocarditis rats
摘要
Abstract
Aim: To establish autoimmune myocarditis rat model and observe the effect of fibrinogen -like protein 2 ( Fgl2) gene overexpression on the heart function of autoimmune myocarditis .Methods:A total of 32 male Lewis rats were randomly allocated into EAM group , EAM-GFP group,EAM-Fgl2 group and control group .A lentiviral vector of Fgl2 gene was constructed.After myosin injection,a trans-thoracic echocardiographic analysis was performed on day 60,and LVFS and LVEF were measured.The expression of Fgl2 was measured by Western blot .BNP level was estimated with ELISA kit for BNP.Results:Compared with control group ,EAM group and EAM-GFP group,the expressions of Fgl2 and BNP were sig-nificantly increased in EAM-Fgl2 group(P<0.001),while LVFS and LVEF were significantly decreased (P<0.001). Conclusion:Fgl2 gene overexpression deteriorates the heart function of rats with EAM .Fgl2 is a potent target for the treat-ment of EAM .关键词
