摘要

Abstract

Objective:In vitro cultivation of SACC-M cells and explore the best concentration of icariin and best point in time on anti SACC-M as well as study the alteration of Bcl-2、Bax protein after SACC-M cells were treated by i-cariin. Method: ①The SACC-M cells after exposure to different concentrations of icariin (0μg/mL、6.25μg/mL、12.5μg/mL、25μg/mL、50μg/mL、100μg/mL、200μg/mL、400μg/mL、800μg/mL)for different time periods (24 h、48 h、72 h) was determined by MTT assay,then explore the best concentration of icariin and best point in time on anti SACC-M cells. And through the morphological observation test ICA's effect on salivary gland adenoid cystic cancer cells apoptosis; ②Compare the protein expression discrepancy of Bax、 Bcl-2 in non- icariin-treat and icariin- treat SACC-M cells which after incubation with best concentrations of icariin and best point in time were detected by using Immunohisto-chemistry. Result:①Compare with the control group(0 μg/mL),icariin in different concentrations(50 μg/mL、100 μg/mL、200μg/mL、400μg/mL、800μg/mL) icariin can inhibit proliferation of cancer cells(P<0.05),compared with oth-ers of 24 h action which were in the same concentration,it was statistically significant from 50μg/ml concentration (P <0.05),but was of no time dependence till it's concentration to the 800 μg/mL concentration(P>0.05).When use(50μg/mL、100 μg/mL、200 μg/mL、400 μg/mL) icariin in cells after 24 h,the ACC-M cells presented typical morphological features of apoptosis by microscope.②Icariin depend on concentration of inducing salivary gland adenoid cystic cancer cells apoptosis,the higher the concentration,the higher apoptosis rate,lower expression of Bcl-2 and rise the expression of Bax. Conclusion:Epimedium glycoside can inhibit the growth of ACC-M in vitro,its mechanism may be related to lower expres-sion of Bcl-2 and rise the expression of Bax.
